EICOSANOIDS AND RELATED COMPOUNDS – AN INTRODUCTION Eicosanoid Structures and Key Enzymes
نویسنده
چکیده
The term eicosanoid is used to embrace biologically active lipid mediators (C20 fatty acids and their metabolites), including prostaglandins, thromboxanes, leukotrienes and other oxygenated derivatives, which are produced primarily by three classes of enzymes, cyclooxygenases (COX-1 and COX-2), lipoxygenases (LOX) and cytochrome P450 mono-oxygenases. The key precursor fatty acids are 8c,11c,14c-eicosatrienoic (dihomo-γ-linolenic or 20:3(n-6)), 5c,8c,11c,14c-eicosatetraenoic (arachidonic or 20:4(n-6)) and 5c,8c,11c,14c,17c-eicosapentaenoic (20:5(n-3) or EPA) acids (see our web page on ‘polyunsaturated fatty acids’). However, it is now impossible to discuss these compounds and their biological activities properly without also considering the docosanoids (resolvins and protectins) derived from 4c,7c,10c,13c,16c,19c-docosahexaenoic acid (22:6(n-3) or DHA), and related products formed by non-enzymic means (isoprostanes). Similarly, plant products such as the jasmonates and other oxylipins derived from 9c,12c,15coctadecatrienoic (α-linolenic or 18:3(n-3)) acid have analogous structures and functions. It is noteworthy that the precursor fatty acids for all of these belong to both the omega-6 and omega-3 families.

متن کاملEnzymic lipoxygenation of arachidonic acid: mechanism, inhibition, and role in eicosanoid biosynthesis
Studies on the lipoxygenation of arachidonic acid by various enzymes have been carried out using diverse approaches. A mechanistic picture of this reaction has been developed which both correlates known information and assists in the rational design of new lipoxygenase inhibitors. A working hypothesis is advanced for the biosynthesis of prostaglandins in mammalian systems. An entirely different...
متن کاملLipid Mediators Are Critical in Resolving Inflammation: A Review of the Emerging Roles of Eicosanoids in Diabetes Mellitus
The biosynthesis pathway of eicosanoids derived from arachidonic acid, such as prostaglandins and leukotrienes, relates to the pathophysiology of diabetes mellitus (DM). A better understanding of how lipid mediators modulate the inflammatory process may help recognize key factors underlying the progression of diabetes complications. Our review presents recent knowledge about eicosanoid synthesi...
متن کاملFunctional eicosanoid test and typing (FET) of peripheral blood cells in eicosanoids related diseases.
Monitoring of eicosanoid synthesis in peripheral blood cells has significant potential for improving the diagnosis and therapy of many human diseases. The quantitative relation between concentrations of prostaglandins and leukotrienes is central to the physiologic function of the eicosanoid network. Here we show that this regulation, which we call the functional eicosanoid typing (FET), fluctua...
متن کاملEicosanoids Derived From Arachidonic Acid and Their Family Prostaglandins and Cyclooxygenase in Psychiatric Disorders
Arachidonic acid (AA)-derived lipid mediators are called eicosanoids. Eicosanoids have emerged as key regulators of a wide variety of physiological responses and pathological processes, and control important cellular processes. AA can be converted into biologically active compounds by metabolism by cyclooxygenases (COX). Beneficial effect of COX-2 inhibitor celecoxib add-on therapy has been rep...
